LOMECAN V® is an Azolic Antifungal with effective activity against Candida albicans, Candida tropicalis, Candida glabrata, C. neoformans, B. dermatitidis, H. capsulatum, C. immitis, Paracoccidioides braziliensis. The drug has antiprotozoal effects against Leishmania major. It is extremely effective in the treatment of vulvovaginal candidiasis.

PHARMACEUTICAL PROPERTIES

PHARMACOKINETICS AND PHARMACODYNAMICS: The absorption of Clotrimazole in the vagina is 3 to 10%. Even three days after administering it vaginally, it persists in fungicidal concentrations. The small amount that is absorbed is metabolized in the liver and excreted in the bile. Clotrimazole is an azole antifungal from the imidazole group whose mechanism of action is the inhibition of sterol 14-α-demethylase in fungi, which is an enzyme system that depends on Cytochrome P450 of microsomes. Thus, imidazoles hinder the biosynthesis of ergosterol in the cytoplasmic membrane and allow the accumulation of 14-α-methylsterols.

These methylsterols alter the close arrangement (packing) of the acyl chains of phospholipids and thereby alter the functions of some enzymatic systems of the membrane, such as ATPase and enzymes of the electrical transport system; They thus inhibit the proliferation of fungi. At the concentrations reached with the local use of Clotrimazole, the permeability of the cytoplasmic membrane of the fungus is directly increased.

Every 100 g contains:

Clotrimazole   2.00g

Excipient c.b.p. 100.00 g.

CONTRAINDICATIONS

The drug is contraindicated in case of hypersensitivity to antifungals of the imidazole group or to any other component of the formula.

DOSAGE AND ROUTE OF ADMINISTRATION:

Route of administration: Vaginal.

Dose:

In vulvovaginitis: Using an applicator, introduce 5 g of 2% cream once a day for 3 days, vaginally.

Application mode: It is recommended to apply at night before going to sleep. The patient, lying on her back with her legs separated and semi-flexed, introduces the applicator containing 5 g of Clotrimazole 2% cream, deposits it and remains in this position for 15 minutes.
